A type of excipient added during the vacuum freeze-drying process to protect bioactive substances from physical and chemical damage.

What is a lyoprotectant?

A lyoprotectant is an excipient added to a formulation before freeze-drying (lyophilization) to protect the active biological or pharmaceutical material from damage during freezing and drying.

The word comes from:

  • Lyophilization = freeze-drying
  • Protectant = substance that protects the product

The basic concept is:

Liquid formulation → freezing → primary drying → secondary drying → stable dry cake/powder → reconstitution

During freezing and drying, proteins, vaccines, enzymes and nanoparticles can experience ice-induced concentration, dehydration, aggregation, denaturation and structural damage. Lyoprotectants help reduce these effects.
